The ammonium acetate in the retort was estimated in the usual way after alkalisation by distilling into 50cc of N sulphuric acid solution.
Experiment had shown that 10cc of morphine solution (the hydrochloride dissolved in water so as to have 0.001 gram of morphine in 1cc), 80cc of water, and 10cc of 25 per cent sodium hydrate aqueous solution, could be subjected to steam distillation without the loss of any morphine, alkaloid being indicated by iodic acid and ammonia in the distillate, and that in the retort by the same test being found to be undiminished in amount. Therefore in the opium smoke experiment the morphine was separated from the original retort contents (from which it has been shown the pyridine bases and ammonia had been removed) by shaking with amyl alcohol, passing into acid water, and estimating with iodic acid and ammonia. This method (George - Analyst 1906 p265) of determining small quantities of morphine in a complex mixture has been found to be trustworthy.
When the solution under examination contains other substances such as sodium chloride, sulphate, hydrochloric or sulphuric acid, it is essential in quantitative experiments for the control solution to have the same quantity of these substances.
